Derwent Biotechnology Resource provides complete coverage of
biotechnology literature and patents worldwide.

DWBA contains information on all aspects of biotechnology including:
genetic engineering, biochemical engineering, pharmaceutical
technology, agriculture, cell culture, biocatalysis, waste disposal
and the environment.

Abstracts for DWBA are specially written by a subject specialist,
and each abstract contains a concise summary of methods, results and
conclusions. All abstracts are in English.

Full bibliographic details are provided including author reprint
address for literature, and full legal information for patents. A
controlled vocabulary is used for easy and accurate searching.
About one third of the database covers patents.

DWBA will be of use to research scientists in biotechnology,

Abstract
Glucose-6-phosphate-dehydrogenase (G6PDH, EC-1.1.1.49) from
Leuconostoc mesenteroides is produced by: isolating genomic DNA
encoding G6PDH from L. mesenteroides; preparing oligonucleotides
complementary to the upstream and downstream DNA of the G6PDH
gene; subjecting the genomic DNA to the polymerase chain reaction
(PCR) with the oligonucleotides; digesting the amplified DNA with
BamHI and SphI to produce a restriction fragment; ligating the
fragment into a plasmid; introducing the plasmid into a suitable
Escherichia coli strain; and isolating G6PDH from a recombinant
E. coli culture. DNA sequences for the primers and the G6PDH gene
are specified. Cloning of the G6PDH gene from L. mesenteroides
allows sequence comparison with the equivalent gene in other
species. Such information is useful in designing site-directed
mutagenesis experiments in order to determine the enzyme
mechanism. In an example, the G6PDH gene was cloned in E. coli
DH5-alpha MCR, using plasmid pUC19 as vector. Recombinant plasmid
plmz, containing the entire gene, was isolated. The specific
activity of G6PDH was 5,000 times increased in the recombinant
strain. (22pp).

4. Output from online searches may be stored temporarily in
electronic form for a maximum period of three months for the
purpose of editing or reformatting the search results. For longer
term storage, a Downloading Agreement with Derwent must be
completed and the appropriate fees paid.

5. In the event of a breach by the customer, its employees or any
third party, of any of these terms and conditions Derwent shall
be entitled to terminate the customer's right of access to
Derwent databases immediately without notice.

6. Derwent shall not in any circumstances be liable or responsible
for the completeness or accuracy of any data comprised in the
Derwent databases nor for the transmission of such data and will
not be liable for any indirect, consequential or economic loss of
profit resulting directly from the use by the customer or any
third party of such data.

7. The customer will not assign any of its rights under these terms
and conditions in whole or in part without the prior written
consent of Derwent.

8. The invalidity, illegality or unenforceability in whole or in
part of any of these terms and conditions shall not affect the
validity, legality and enforceability of the remaining terms and
conditions.

9. These terms and conditions shall override and prevail over any
other terms and conditions in any document or other communication
used by the customer in effecting its subscription to the Derwent
databases.

10. These terms and conditions shall be governed by and construed in
accordance with English law.
